Isolation of Microfibrils from Rat Vibrissal Follicles
نویسنده
چکیده
The fibrillar nature of wool keratin is well established but considerable doubt still exists concerning the mode of aggregation of the constituent protein molecules . Early workers (7, 10, 12, 22) showed that it is possible to examine the contents of cortical cells of suitably treated wool samples with the electron microscope, and that (macro)fibrils, which can be discerned in the optical microscope (10), consist of smaller filaments with diameter ranging from 500 nm down to about 10 nm . Farrant et al. (7) considered the microfibril to consist of globular keratin molecules (10 nm in diameter) but it was pointed out by Jeffrey et al . (12) that "sufficiently drastic treatments (of wool) are effective in producing a globular appearance in the microfibrils" and they observed that the more complete isolation of microfibrillar material required extreme breakdown of both hydrogen bonds and disulphide cross-linkages . However, with the advent of ultramicrotomy and the application of electron stains such as osmium tetroxide (1, 19), thin cross-sections of wool revealed the typical filament-matrix texture, which consists of microfibrils, 7-8 nm in diameter, embedded in an osmiophilic matrix (19) . High resolution micrographs show microfibrils to possess a fine structure which Filshie and Rogers

A quantitative study of hair growth using mouse and rat vibrissal follicles. I. Dermal papilla volume determines hair volume.
The volumes of the dermal papilla, matrix and fully grown hair of vibrissal follicles of rats and mice were measured. Dermal papillae were surgically removed from rats' vibrissal follicles and a small new dermal papilla was always reformed. The lower third of rats and mice follicles, containing the whole of the dermal papilla was amputated and very small dermal papillae were reformed. Follicles...

متن کاملA mechanism of rat vibrissal movement based on actual morphology of the intrinsic muscle using three-dimensional reconstruction.
The vibrissal capsular muscle (VCM) of the rat is known to differ from the arrector pili muscle. The purpose of the present study was to characterize the rat VCM morphologically using three-dimensional reconstruction. The rat snout skin was fixed, processed with routine histological methods, sectioned serially at a thickness of 10 µm, and then stained with Masson's trichrome. The sectioned imag...
متن کاملDendritic pattern in mouse barrel field after a neonatal vibrissal follicles removal: MAP-2 immunohistochemistry.
A distribution of dendrites was studied in mouse barrel field after a neonatal unilateral partial lesion of vibrissal follicles using anti-MAP-2 immunohistochemistry. The effect of a neonatal vibrissal follicles removal was studied in adult mice: barrels corresponding to intact follicles were enlarged whereas those representing removed follicles had not developed. MAP-2 immunopositive profiles ...
متن کاملLocalization of Myosin and Actin in the Pelage and Whisker Hair Follicles of Rat
The combined effects of myosin II and actin enable muscle and nonmuscle cells to generate forces required for muscle contraction, cell division, cell migration, cellular morphological changes, the maintenance of cellular tension and polarity, and so on. However, except for the case of muscle contraction, the details are poorly understood. We focus on nonmuscle myosin and actin in the formation ...
